In the film, set in a fictional 1947 Hollywood, cartoon characters (called "Toons") live and work alongside real human actors in a seemingly peaceful coexistence. Roger Rabbit is a famous Toon actor known for his zany antics and his marriage to the sultry Jessica Rabbit, who works as a singer and performer at a popular nightclub.

When Roger becomes convinced that Jessica is cheating on him, he seeks the help of Eddie Valiant, a private detective who specializes in cases involving Toons. With the police hot on his trail, Roger turns to Eddie for help in clearing his name and finding the true killer. Along the way, the unlikely duo encounters a colorful cast of characters, both human and Toon, including the villainous Judge Doom, who is determined to rid Toontown of its Toon population once and for all.
