The film begins with Tweety attending a meeting at the Looney Club, where various Looney Tunes characters gather to discuss different topics. Colonel Rimfire, a hunter and an arrogant cat, claims that cats are the most intelligent animals and can easily outsmart their prey. Granny, who runs the club, disagrees and believes that Tweety is the smartest animal, which leads to an argument between Rimfire and Granny.

To settle the debate, Granny proposes a bet: Tweety must fly around the world in 80 days and collect the pawprints of 80 cats from different countries. If Tweety succeeds, Granny will win enough money to save a local park from being demolished. If Tweety fails, Rimfire gets to keep the club.

Sylvester, Tweety's longtime nemesis, overhears the bet and decides to follow Tweety around the world. He plans to catch Tweety and eat him, but he also wants to collect the pawprints himself to win the bet for Rimfire. A chase begins, and the two animals travel to various locations, including Paris, China, and Africa.

Along the way, they encounter other Looney Tunes characters, such as Bugs Bunny, Daffy Duck, and Yosemite Sam. Some of them help Tweety, while others try to hinder him. Meanwhile, Granny anxiously waits for updates on Tweety's progress.

Eventually, Tweety and Sylvester make it back to the Looney Club within the 80-day deadline, and the pawprints are counted. The reveal of who wins the bet is left ambiguous, but Rimfire concedes that cats may not be the smartest animals after all. Tweety and Sylvester settle their differences and finally end up as friends.
