Michael Dorsey, played by Dustin Hoffman, is a talented but difficult-to-work-with actor in New York City who is constantly struggling to find work. When he learns that his friend and former roommate Jeff, played by Bill Murray, has landed a role on a popular daytime soap opera, Michael decides to audition for a part as well. However, he quickly realizes that the producers aren't interested in hiring him because of his reputation for being difficult.
In a moment of desperation, Michael comes up with a plan: he'll create a female persona named Dorothy Michaels and audition for the part in character. Much to his surprise, he gets the role and becomes an instant hit with viewers. As Dorothy, Michael learns how differently women are treated in the entertainment industry and finds himself increasingly drawn to his co-star, Julie, played by Jessica Lange.
