The three actors, Lucky Day (Steve Martin), Dusty Bottoms (Chevy Chase), and Ned Nederlander (Martin Short), are struggling to find work in Hollywood, when they receive an invitation from a small Mexican village to come and perform as their legendary hero characters, the Three Amigos. Mistakenly believing the invitation to be for a film shoot, the three eagerly accept and head to the village.
Upon their arrival, they are quickly informed that the villagers were expecting them to actually fight off a real bandit, El Guapo (Alfonso Arau), who has been terrorizing the town. The Three Amigos are initially reluctant to take part, but they ultimately agree to help the villagers.
As they prepare for the inevitable confrontation with El Guapo and his gang, the Three Amigos begin to realize that they are not as skilled in fighting as they thought. However, with the help of the village residents and their own comedic ingenuity, they manage to outsmart and defeat El Guapo and his band.
In the end, the Three Amigos are hailed as heroes and decide to stay in the village to help the residents rebuild and prepare for any future threats. The film ends on a humorous note with the Three Amigos performing a musical number and the villagers joining in.
