Jake Blues is released from prison and picked up by his brother Elwood in a battered old police car that they borrowed from their former nun caretaker, Sister Mary Stigmata. Upon arrival at the orphanage where they grew up, the brothers discover that it is in dire financial straits and will soon be closed down unless $5,000 is raised.

In an attempt to save the orphanage, Jake and Elwood set out on a mission to raise the necessary funds. They decide to reform their old rhythm and blues band by collecting members from various establishments around Chicago, including a country bar and a soul food restaurant.

As they make their way around the city, playing their music and collecting money, the brothers also find themselves in a number of sticky situations. They are chased by the police, pursued by a vengeful ex-girlfriend, and even hounded by a neo-Nazi group. Meanwhile, a vengeful woman named Carrie Fisher, who plays the character of Jake’s girlfriend, is pursuing them.
