After being separated from the Straw Hat Pirates, Chopper is initially terrified on the island but is soon befriended by the animals who see him as their savior. They believe him to be the king they have been waiting for to protect them from the "horn" hunters who have been terrorizing them. Chopper is hesitant to accept their praise but slowly starts to believe in himself.
Meanwhile, Luffy and the rest of the crew come across Momambi, a young boy who despises pirates due to his past experiences with them. Despite their initial animosity towards each other, they soon team up to find their missing crewmate and stop the hunters from harming the animals.
As they make their way through the island, they also learn about the legend of the island's treasure and its power to grant immense strength to whoever possesses it. Count Butler, the leader of the hunters, is obsessed with obtaining this power and plans to use it for his own gain.
As the two groups finally come face to face, a battle ensues between the Straw Hat Pirates and the hunters. Chopper realizes that he must use his medical skills to help his friends and the animals, and becomes a vital member of the fight.
In the end, the Straw Hat Pirates are able to defeat the hunters and save the island's animals. Momambi also learns that not all pirates are bad and begins to trust Luffy and his crew. The film ends with the crew sailing off to continue their adventures through the Grand Line.
